WDRs are pivotal protein-protein interaction hubs that play roles in a wide range of cellular processes. SGC has previously demonstrated that WDRs are druggable, and the exceptional disease association of this target class is a call for increased efforts to develop small-molecule WDR antagonists. However, compared to other prominent drug target families, a majority of human WDRs remain largely uncharted. This month, Levon Halabelian's group has released several WDR structures in complex with inhibitors, notably WDR5 and WDR91. These compounds were identified using DEL-ML, in collaboration with Relay Therapeutics, X-Chem, Inc., and Google. Two months ago we shared here our (with Gabriela Teixeira) first case of pDVA in a no-option CLTI patient. Here is the follow-up. One week after treatment , we detected thrombosis of the graft. We performed a successful thrombectomy with 6F Rotarex, following the suggestion from Bruno Migliara. On completion angio, a tight stenosis of the last viabahn (under the retinaculum) was visible and was treated with high pressure balloon angioplasty. Now, after more than one month working well, we keep watching the gangrene delimitation. #CLTI #endovascular #vascularsugery #pdva

Found it interesting as when we were being trained on the effects of SGLT2is when I came onboard in 2013, we were trained on the differences in the diameters between the afferent and efferent arterioles, these differences resulted in the hemodynamic changes seen with: *RAASis *SGLT2is *nsMRAS *other agents impacting the neurohormonal axis. AdAlta Ltd CEO and Managing Director Dr Tim Oldham spoke to Proactive about the results of AD-214 dose simulation studies that support the potential efficacy of the planned intravenous dose regimen for treating fibrotic disease.   Dr Oldham notes that this is the most significant data AdAlta has received this year, helping to determine a dose and dosing regimen ahead of Phase II, that is likely to be both efficacious and suitable for clinician and patient adoption. He explains that the model of AD-214 distribution and receptor blocking is a big move forward in de-risking Phase II studies for partners.   The model also allows the exploration of enhanced routes of administration, and suggests the potential for more convenient subcutaneous administration. This could be available in time for Phase III if a partner wishes to progress this option.   This data is exciting news for AdAlta and adds real momentum to its partnering program for both strategic pharma opportunities and project financing options going forward.
