Check our last publication about coupling high throughput matrix-compatible SPME with automated Microfluidic open interface (MOI) for whole blood analysis! Fentanyl analogues are mainly synthesized to evade analytical detection or increase their potency; thus, very low concentrations are sufficient to achieve a therapeutic effect. By using Concept96 SPME autosampler with Shimadzu 8060 via MOI interface, our quantitative method is capable of extremely sensitive analysis and rapid analysis (2.4 min per sample), with limits of quantification ranging from 0.002 to 0.031 ng mL–1 and linearity ranging from 0.010 to 25.0 ng mL–1. The method shows very good reproducibility (1–18%), accuracy (81–100%) of calibration and validation points, and good interday reproducibility (6–15%). Check out the recent paper in Small where we show palladium-catalyzed Suzuki–Miyaura reaction at the nanoliter scale on droplet microarrays for the synthesis of an 800-compound library of biphenyls. Each reaction is carried out in individual 150 nL droplets. Remarkably, the synthesis of these 800 compounds requires only 80 µmol of reagents, and 400 µL of solvent. Via the solid-phase synthesis approach, the compounds could be purified prior to the analysis, while a UV-cleavable linker allows for fast cleavage of each compound into the individual 100 nL droplets. This novel approach expands the toolbox of the droplet microarray for miniaturized high-throughput chemical synthesis and paves the way for future synthesis and screening of chemical compounds in a single platform.
