Seed VCs are turning to new ‘pro rata’ funds that help them compete with the big firms Alpha Partners, SignalRank and now SaaS Ventures help seed VCs pay for shares when big VCs try to price — or push — them out Lee Edwards, partner at Root VC, has a saying at his firm that “pro rata rights are earned, not given.” That may be a bit of a stretch since pro rata refers to a term that VCs put in their term sheets that gives them the right to buy more shares in a portfolio company during consequent funding rounds to maintain an ownership percentage and avoid dilution. Still, while these rights are not exactly “earned,” they can be expensive. The latest update from the research team at Primal Capital explains the importance of wallets in the Web3 ecosystem and highlights the evolution of solutions in the market today. These solutions provide greater utility compared to the original Web3 wallets, which was simply to safeguard assets from centralized platforms.

The GLP-1 space has continued to explode, with the potential for impact expanding to cardiovascular risk, sleep apnea, and liver disease. So who are the emerging players poised to challenge Lilly and Novo? The incoming players to watch are a mix of startups and incumbents. In the startup realm, Viking Therapeutics, Inc., and Altimmune, Inc. have gained significant attention as their GLP-1 candidates demonstrate solid weight-loss outcomes. For San Diego based Viking, weekly doses of VK2735 delivered weight loss of up to 13% compared to placebo in a Phase 2 trial. Perhaps most exciting, the weight loss did not plateau at any time during the 13 week study, suggesting longer duration doses would result in further weight loss. Meanwhile, Altimmune's Pemvidutide (being studied for both obesity and MASH) is on track for an end of phase 2 FDA meeting in Q3 of this year. Notably, Pemvidutide has showed promise in limiting the amount of weight loss derived from muscle, which is a notable problem with the existing GLP-1s. A recent phase 2 trial showed only ~25% of weight loss was from lean mass, more in line with the ratio seen in typical diet & exercise programs. There are also several incumbents working hard to establish their presence. Roche's $2.7B acquisition of California-based Carmot Therapeutics, Inc. Therapeutics in December and Amgen's MariTide are both notable. Roche recently announced data from a Phase 1B study for CT-388 showing placebo-adjusted weight loss of ~19%, with 85% of participants seeing weight loss of more than 10%. Not to be left out, Amgen has recently advanced MariTide into Phase 2 studies, with Phase 1 results published in February showing weight loss of up to 15% in 12 weeks, with some dosage levels only required once a month. To have a credible chance at challenging the existing market leaders, new GLP-1s will have to demonstrate meaningful improvements on the following: 1. Dosage - most approved therapies today (with the exception of Rybelsus) are subcutaneous injections, given weekly. Having an easier form (tablet) or frequency (bi-weekly or even monthly) would be a big advantage. 2. Preservation of lean muscle mass - helping patients derive weight gain from regular adipose tissue instead of muscle mass. 3. Benefits beyond weight loss. Novo's recent FDA label expansion for Wegovy for reducing the risk of serious cardiovascular events provides a blueprint of how to expand the area these medicines can be helpful in. While there are many companies active in the space, it's worth noting just how large a lead Lilly and Novo have in this future $100B+ market. A recent GlobalData report noted that even by 2029, these two giants will have ~80% of the market between them. That said, 20% of a $100B market is still a sizable prize for other innovators, and continued innovation will only help patients.

Interesting piece from Marc Andreessen on how startups are not getting the regulatory support they need to sustain America's technological leadership. "Regulatory agencies have been green lit to use brute force investigations, prosecutions, intimidation, and threats to hobble new industries, such as Blockchain. Regulatory agencies are being green lit in real time to do the same to Artificial Intelligence." While many investors themselves (both in the US and Europe) are worried how AI will change our lives, our jobs and social interactions, the top VC opinion leader thinks regulation is already too restrictive. #techstartups #techleadership #strategicgrowth

Ross Haleliuk posted the fantastic article below. This is the world where I live day-to-day. At TechOperators, we invest mostly in early-stage cyber, but we do things a little differently, and we believe there are ways to invest successfully outside of pure Power Law math. The article argues that many security problems are too small for VC. I agree. I often try to convince bootstrapped founders not to raise venture because doing so can turn a successful, slow-growing bootstrapped company into a failed venture-backed company because, despite a large infusion of capital, it couldn't double every year. VC is not monolithic--not by stage, strategy, or style. Venture is often equated with "Tier 1 Venture". Ross argues that VC is not always great for early-stage cyber--and he is right. Bootstrapping AND VC work when incentives are aligned. Does it work for an early-stage VC with a <$200M fund to invest in several early companies at reasonable valuations, setting up the conditions for reasonable exits that pay off for both investors and founders? Yes. Does it work for $800M funds investing in seed stage at $100M+ valuations? Well, that depends! Power law says it does (for VC), but the unfortunate externality is that these rounds destroy companies and founder equity more often than not. There is a role for patient capital in this ecosystem to fuel successful companies that retain exit optionality as they scale--driving exit value for both founders and investors.

The opioid crisis was a sad hallmark of my years working for the Baker Administration. In a time of extreme partisanship at the federal level, this was one area where public policy, civil discourse, and private enterprise seemed aligned and sufficiently motivated to find a solution. What we learned, however, is that dollars and access only go so far in combatting a disease that knows no so specific demographic. Instead, it's apparent that a battery of tools is needed to combat and prevent addiction--ranging from strong safety nets, to novel non-addictive pain therapies, and better technology. Introducing Marigold Health. Shrenik Jain, Ravi Shah, Tracey Cohen, MD, FASAM and team have built a proprietary NLP-based platform that--most significantly--improves the lives of those suffering from SUD/SMI. One customer we spoke with noted, "We needed a unique treatment intervention that had supportive research, fit into a CMS established model with a recognized service code, and that would provide positive clinical outcomes in an efficacious and fiscally responsible manner”. Another plainly said: "Marigold is our best hope to reach into the lives and help those who many have already given up on." Now you can sequence your every cell one by one. Expect that for protein folding (AlphaFold was that moonshot) and other simulation uses, a full "twin" model of you, cell by cell -- Google has no angle here -- worse chips, worse cloud, worse models, worse phones / edge devices -- Specialty silicon for unique use-cases, lower-power, lower-memory are surely coming fast - like back in the day for mobile phones when Qualcomm, Broadcomm, Infineon, Mediatek, etc duked it out.

I’m optimistic about the potential of generative AI to reduce provider burnout and improve provider-patient interactions. However, that optimism is tempered by the data that indicates the technology is not quite ready yet. Bright Ventures is interested in digital health founders who understand the realities of provider-patient interactions, and the role technology can play in strengthening clinical outcomes. This is a good resource I've stumbled upon. It covers a plethora of VC and startup-related topics and contains links to expand your knowledge. https://lnkd.in/dqkdV4fA TL;DR: "It's the founders, stupid!" These two lines made me hover over: "I know the physical and emotional tolls that entrepreneurs face". "Resources for Managing Founder Anxiety and Depression" I've been one myself, and it's not for the faint-hearted. I struggled and failed many times. From the standpoint of a person working now for a VC fund, the ability to support a founder who was hit with too much at the same time, to know how to listen, mirror and label Chris Voss's style, and a bit of guidance to build confidence back can do wonders. Assisting portfolio companies doesn't have to be just intros and throwing money or unsolicited advice. And what about identifying troublesome founders before investing to prevent assisting portfolio companies from becoming a nightmare flop? Ahh, that's a tricky one. Startups fail for a variety of reasons. At the early stage, we should look at many things, to name a few: product-market fit, go-to-market, blue ocean or zero to one, CAC and LTV ratio, CAP table, passion, leadership potential, intelligence, technical ability, and many, many more. Crunch the numbers. Sure. What about the early stage? Limited information and knowledge when a decision has to be made, and the outcome is impossible to predict because the future is unknown and unknowable. Examine the founders. If you don't read the founders right and overlook some hidden toxic traits and coping mechanisms that mask something that can derail the startup and founders down the road (vulnerable narcissism, antisocial tendencies, Machiavellism, blameshifting or more benign like anxiety-prone, conflict avoidance, burn-out tendency, inability to delegate, perfectionism, risk aversion, etc.), the chances that it will be your next outlier significantly diminish. How many of you saw dead equity on the capable, founders' clash, low morale, a founder playing the sunk cost game with the fund after burning cash, lying, or even more brazen behaviours like experimenting with horse tranquilisers and smoking weed on a podcast? A "Sudden" shift of values with a splash of megalomania could lead to a CEO removal attempt by the board. Or stealing "HER" voice. It is fun to watch as a TV drama, not to deal with as your portfolio. We won't be able to see it all during a few conversations with the founders and not everyone is Wendy Rhoades from Billions. Knowing how to ask a question behind which is a hidden question and being able to detect when founders say what they think we want to hear to get the funding is a good start. Spending extra time learning that is a good investment. A splash of psychology, empathy, self-awareness, and emotional intelligence to become an excellent GP won't harm your ability to identify and help the right entrepreneurs build great companies.
