Research recently published by the Chase Beisel Lab showcases the remarkable potential of reprogrammed tracrRNAs from diverse Cas12 nucleases, allowing the detection of RNA with dsDNA cleavage and collateral single-stranded DNA cleavage, all without the RNA necessarily encoding a protospacer-adjacent motif (PAM). The implications of this work extend to diverse dsDNA-targeting Cas12 nucleases, significantly expanding the flexibility and versatility of CRISPR-based RNA detection.
